Transforming local cancer treatment through smart drug retention

We are developing a differentiated 505(b)(2) product for the local treatment of a difficult-to-manage urothelial cancer affecting the upper urinary tract. The programme combines a well-established anticancer agent with a temperature-responsive delivery system designed to improve drug retention at the site of disease.

This matters because conventional liquid instillations may be rapidly cleared from the urinary tract, limiting contact time between the medicine and the tumour. Patients may therefore require repeated procedures, and some may ultimately face major surgery with significant consequences for renal function and quality of life. Our objective is to create a practical, organ-preserving treatment option that delivers high local exposure while limiting unnecessary systemic exposure.

The product is administered as a liquid through a catheter. At body temperature, the formulation transitions into a semi-solid depot, allowing the active drug to remain in contact with the urothelial surface for an extended period. The depot then gradually dissolves and is eliminated naturally through urinary flow. This localised delivery approach is intended to improve residence time, treatment consistency, and clinical usability.

The underlying therapeutic concept has already been clinically validated through an approved product in the same disease setting. Our programme builds on that regulatory and clinical precedent while pursuing a differentiated formulation, manufacturing process, intellectual property position, and commercial profile.

Work completed to date includes product-concept definition, target product profile development, preliminary formulation assessment, excipient and material selection, patent-landscape review, regulatory pathway analysis, and development-risk evaluation.

The next phase will focus on , in-vitro performance testing, stability studies, process scale-up, container-closure selection, GMP batch manufacture, and regulatory engagement.
