Chromatography-free purification methodology
Airobes developed and validated a fusion protein architecture that enables (i) chromatography-free purification to minimum purities of 85-90%, (ii) reduced cytotoxicity and aggregation for difficult-to-express and intrinsically disordered proteins/peptides, and (iii) high-throughput screening of generative AI protein designs for foldability in vivo. The method shortens post-fermentation processing to at total of 3 hours and eliminates the need for chromatography, while maintaining industrial-grade protein purity. The protein of interest is fused to a proprietary transmembrane anchor, allowing the membrane to be used as a solid support for subsequent wash steps - akin to chromatography resin. The target protein is released via digestion with a site-specific protease. A provisional U.S. patent has been filed.
